What parameters to setup in Jmeter

I have a scenario where:

For every 30 seconds 10,000 users will hit the server with a difference of 0.5 seconds (approx) between each user's request, and this test should be done for 1 hour.

Now How do i set up all the JMeter parameters to achieve this.

When should i use more than one Thread groups?
